چکیده

Accepted 13 May 2013 Perillyl alcohol and d-limonene are naturally occurring plant compounds that exhibited anticarcinogenic activities in mammary tumor models. The effects of these monoterpenes at the initiation stage of aflatoxin B1-induced hepatocarcinogenesis were investigated. Male F344 rats were fed Control or treatment diets throughout the study and exposed to aflatoxin for 5 days. Three days after the last aflatoxin dose, blood and liver samples were obtained. Analysis of liver samples showed that both limonene and perillyl alcohol significantly inhibited (p<0.05) aflatoxin-DNA adducts formation in hepatocytes. The monoterpenes may have potential for use as chemopreventive agent against aflatoxin-induced liver cancer.

Predictive value of molecular dosimetry: individual versus group effects of oltipraz on aflatoxin-albumin adducts and risk of liver cancer.

Studies in animals and humans have established serum aflatoxin-albumin adducts as biomarkers of exposure to aflatoxin B1 (AFB1), a food-borne hepatocarcinogen. To assess the utility of measurements of aflatoxin-albumin adducts to predict risk of hepatocellular carcinoma (HCC), 123 male F344 rats were dosed with 20 microg of AFB1 daily for 5 weeks after randomization into three groups: no interv...

Quantitation of Aflatoxin B,-modified DMA Using Monoclonal Antibodies1

Five monoclonal antibodies were obtained after fusion of mouse P3 x 63 myeloma cells with spleen cells isolated from BALB/c mice that had been immunized with aflatoxin B,-adducted DMA complexed with methylated bovine serum albumin. Selected hybridomas were found to produce monoclonal anti bodies specific for modified DMA containing both the 2,3dihydro-2-(/V7-guanyl)-3-hydroxyaflatoxin B, and th...

The formation of AFB(1)-macromolecular adducts in rats and humans at dietary levels of exposure.

The levels of aflatoxin B(1)-DNA and aflatoxin B(1)-albumin adducts were investigated by accelerator mass spectrometry (AMS) in humans and rats following exposure to a known, dietary relevant amount of carbon-14 labeled aflatoxin B(1) ([(14)C]AFB(1)). The aims of the study were to: (a) investigate the dose-dependent formation of DNA and protein adducts at very low doses of AFB(1) (0.16 ng/kg-12...

Quantitation of aflatoxin B1-DNA adducts in woodchuck hepatocytes and rat liver tissue by indirect immunofluorescence analysis.

A quantitative indirect immunofluorescence technique was developed utilizing a monoclonal antibody (6A10) recognizing the imidazole ring-opened form of the major N-7 guanine adduct of aflatoxin B1 (AFB1). This method was used to investigate adduct formation in woodchuck hepatocytes treated in culture and in liver tissue of rats treated i.p. with AFB1. Fluorescein isothiocyanate-labeled secondar...

Aflatoxin B1-2,3-oxide as a probable intermediate in the covalent binding of aflatoxins B1 and B2 to rat liver DNA and ribosomal RNA in vivo.

Administration of[3H]aflatoxin B2 (2,3-dihydroaflatoxin B1)(AFB2) to male rats resulted in levels of hepatic DNA- and ribosomal(r)RNA-aflatoxin adducts that were about 1% of those for rats given [3H]aflatoxin B1(AFB1). The levels of hepatic protein-aflatoxin adducts were 35 to 70% as great for AFB2-treated as compared to AFB1-treated rats...
